The application of differential display of mRNA technique in transgenic mice liver tumorigenesis

Abstract
Tumorigenesis involves multiple genetic aberrations including oncogene activation followed by the potential secondary events occurring as the gain of function (oncogene activation) or the loss of function (inactivation of tumor suppressor gene). In order to identify the genes involved in Hepatocellular carcinoma, the newly developed technique called differential display of mRNA technique was applied to the transgenic mice model system. The transgenic mice develops liver tumor by the liver specific expression of the foreign oncogene (SV40 T antigen) with stage specific changes. 20 different primer set were applied to compare the gene expression patterns of liver RNA from non-tumorous stages (normal as well as preneoplastic) with that of tumors. The multiple up and down-regulated genes were detected in this transgenic mice tumorigenesis. The partial cDNA sequences were determined and reveal the unknown genes as well as known genes including cyclin A gene.
